Innovative Solution Overview
We have achieved breakthrough in Jawbone Regeneration, particularly, a novel Immunomodulatory Therapy to prevent Medication-Related Osteonecrosis of the Jaw (MRONJ).
We are investigating an ultrasound-responsive piezoelectric scaffold that converts external ultrasound waves into localized electrical stimulation. This bioelectrical signaling promotes bone regeneration through two complementary mechanisms: Enhancing osteogenesis by activating cellular pathways that stimulate bone-forming cells.Modulating immune responses by encouraging macrophages to shift from a pro-inflammatory (M1) state to a regenerative, anti-inflammatory (M2) state.Unlike conventional bone grafts, membranes, or growth-factor-based therapies, this platform actively directs both immune regulation and tissue regeneration without requiring additional surgery, implanted electronics, or high-dose biologics.
Translating Innovation to Clinical Practice
Our team is the first in Singapore to focus on the prevention of MRONJ through immunomodulatory regenerative technologies. We aim to apply this approach to tooth extraction sites, where MRONJ most commonly develops, by creating a regenerative environment that supports both wound healing and new bone formation . To enhance clinical relevance, we are evaluating this technology in a bisphosphonate-treated minipig model that closely mimics the challenges faced by patients at risk of MRONJ. This represents the first integration of remotely activated piezoelectric scaffolds in a large-animal MRONJ model. By restoring the balance between immune function and bone regeneration, our platform has the potential to transform the prevention of MRONJ and establish a safer, more effective strategy for oral and maxillofacial reconstruction.
A Novel Drug Delivery Hydrogel: Sulfasalazine-Loaded Hydrogels
Our innovative therapy utilizes a biodegradable and biocompatible chitosan methacrylate (CsMe)-based hydrogel as a socket dressing to deliver Sulfasalazine (SAS) directly to the extraction site. Sulfasalazine is an FDA-approved drug that acts as a potent NF-κB inhibitor. By suppressing the NF-κB signaling pathway, the hydrogel:Regulates the Immune Response: It controls the expression of inflammatory genes and cytokines like TNF-α and IL-1β.Promotes Regeneration: It mitigates inflammation-associated necrosis, facilitating both wound healing and alveolar bone regeneration. Targets the Microenvironment: Unlike general treatments, this system is specifically designed to address the unique inflammatory environment of extraction sockets in patients on anti-resorptive medications.
